检测体外中性粒细胞自发性激活的方法学研究

  摘要 从细胞形态学和细胞酶组织化学的角度出发,用伪足法和NBT法检测体外中性粒细胞的自发性激活,研究了正常人中性粒细胞体外自发性激活的时相特征,对这两种方法进行了对比分析。发现:(1)检测体外中性粒细胞自发性激活,伪足法较NBT法更灵敏、可靠、且可实时观察;(2)体外中性粒细胞自发性激活的百分率呈现由低升高、再逐渐降低的时相过程,最高激活百分率出现在采血后第3小时。研究结果为今后深入研究中性粒细胞激活与多种疾病的关系提供了方法学基础。

Study of Methods for Measuring Spontaneous

  Activation of Neutrophils in Vitro

  Abstract From the viewpoints of cellular morphology and enzymic histochemistry, we adopted the pseudopod method and NBT test for the measurement of spontaneous activation of neutrophils in vitro The time course curves of spontaneous activation of neutrophils from healthy donor were drawn. After comparing the pseudopod method with NBT test, we conclude that pseudopod method is more sensitive and reliable than NBT test and it can be used for real time observation; and that the curves of time course ascend from lower level to the highest level within 3 hours after the samples are collected, and then they descend gradually. The methodology of this study is available for use in future studies on the relatioship between activation of neutrophils and many diseases.

   2 材料和方法

  2.1 标本的采集

  上午10时采集健康男性青年志愿者肘前静脉血10 ml,共27例。肝素(200 IU)抗凝,分为二份,分别用两种方法检测激活的中性粒细胞的百分率。
