口腔白斑(oral leukoplakia, OLK)为国际公认的癌前病变,其患病率和癌变率分别高达4.9%~48.9%及3.4%~19.8%,是口腔鳞癌的重要病源之一。Ha-ras癌基因在口腔癌的发生中有重要作用,有研究发现口腔鳞癌标本中 ha-ras基因产物p21有高表达[1]。我们采用聚合酶链反应-单链构象多态性分析(polymerase chain reaction-single-stranded conformational polymorphism,PCR-SSCP)技术,对具有多种生物活性的中药绞股蓝(Gynostemma pentaphyllum Mak, GP)[2,3]干预的金地鼠颊囊白斑模型进行动态研究,检测OLK癌变过程中Ha-ras癌基因含第61位密码子突变的情况,探讨GP对癌基因激活的影响及其防癌、抑癌的分子机理。
