胃癌nm23基因mRNA原位杂交和免疫组化研究

　　【摘要】　目的：探讨胃癌mn23-H₁基因表达与淋巴结转移倾向的相关性。方法：采用地高辛标记cDNAnm23-H₁探针，原位杂交检测21例胃癌及癌旁组织；应用单克隆抗体(NM301)SP法检测131例胃癌及60例非胃癌胃粘膜nm23/NDPK表达。结果：原位杂交阳性率57.1%(12/21)；免疫组化阳性率为87.8%(115/131)，其中非转移组阳性率为97.1%(34/35)，转移组阳性率84.4%(81/96)，P＜0.05。结论：胃癌nm23-H₁基因表达与淋巴结转移倾向呈负相关，与PCNA分级呈正相关。

In situ hybridization and immunohistochemical analysis of expression of nm23-H₁mRNA and its productions in human gastric carcinomas

　　【Abstract】　Objective:To elucidate the relationship between expression of nm23 gene in human gastric carcinomas and lymph node metastatic potential.Methods:Twenty-one gastric carcinoma tissues and correponding non-cancer mucosae were examined by in situ hybridization with digoxigenin-labeled nm23-H₁cDNA probes.Immunohistochemical staining was made by the Streptoavidin-Biotin method(SP) with mAb nm23/NDPK,131 gastric carcinoma tissues and 60 specimens of non-cancer gastric mucosae were examined.Results:Twelve(57.1%)out of 21 gastric carcinomas expressed higher levels of nm23mRNA than corresponding non-cancer mucosae.One hundred fifteen(87.8%)out of 131 gastric carcinomas exhibited possitive immunoreactivity for nm23/NDPK,97.1%(34/35)and 84.4%(81/96)were immuno-reactivity for non-metastatic groups or metastatic groups,respectively.Statistics analysis was made using the χ² test,the P value was less than 0.05.Conclusions:Expression of nm23/NDPK in gastric carcinomas was inversely associated with lymph node metastatic potential,directively with PCNA labeling index.
