Relation between shared epitope of HLA-DR antigen β chain and rheumatoid arthritis

【Abstract】 Objective To explore the association between expression of HLA-DR antigen shared epitope (SE) and rheumatoid arthritis (RA) in Han nationality in Wuhan of China.Method The method of DNA amplification with sequence-specific primers (PCR-SSP) was used to dertermine HLA-DRB101,04 genotype in 78 patients with RA and 126 healthy controls in Han nationality from Wuhan.Results There was a significant increase in the presence of amino acid sequence QKRAA of QRRAA both in RA patients overall compared with healthy group (43.6% vs 15.1%,P0.01).RA patients with SE were more severe in tender joint numbers,radiological severity and extraarticular disease than those with SE negative.Conclusion The results suggest that strong association between SE of HLA-DR antigen and susceptibility and severity to RA in Han nationality in Wuhan,and SE may be a useful marker for disease severity and prognosis in the patients with RA.
