【摘要】 目的 探讨老年吸烟者肺癌H-ras基因变化的意义。 方法 对33例吸烟肺癌和22例不吸烟肺癌患者采用纤维支气管镜毛刷刷取支气管脱落细胞,应用PCR-RFLP方法进行H-ras基因突变检测。 结果 肺癌中H-ras基因总突变率为52.7%(29/55),其中吸烟组肺癌H-ras基因突变率为66.7%,不吸烟组肺癌为31.8%,二者差异有显著性(P<0.05),吸烟组肺鳞癌患者H-ras基因突变率为70.0%,明显高于不吸烟组肺鳞癌患者的16.7%(P<0.05)。 结论 提示H-ras基因可能是被烟草激活的一个特殊位点,吸烟是导致H-ras基因突变的重要因素。
Detection of H-ras oncogene mutation in elderly smokers with lung cancers

【Abstract】 Objective To study the relationship between the H-ras oncogene and lung cancers in elderly smokers. Methods The samples were collected from 55 consecutive patients with tissue diagnosed lung cancers by fiberoptic bronchoscopic brushing. Mutation in coden 12 of H-ras oncogene was detected in the exfoliated cells using PCR-RFLP method (polymerase chain reaction-restriction fragment length polymorphisms). Results H-ras oncogene mutation was present in 52.7%(29/55)of total patients with 70.0%(23/33) of smokers and 31.8%(7/22) of nonsmokers (P<0.01).Subgroup analysis showed that H-ras oncogene mutation was present in 75.0%(15/20) of smokers with squamous cell lung cancer as compared to only 16.7%(1/6) of nonsmokers with squamous cell lung cancer (P>0.05). Conclusions Our results suggest that coden 12 of the H-ras oncogene may be a specific target for the mutagenic activation by tobacco smoke, and smoking is an important factor in the induction of H-ras oncogene mutation.
